When you go to ION Desk > Enterprise Locations, what's the status column read as for your new location? If the EC was able to successfully establish connection and communicate with Infor OS in the cloud, then you should see an OK. If it's blank, then I don't believe it's managed to establish connectivity while if it's in ERROR, there could be a health or configuration issue that's terminated or preventing connection.
Have you checked to ensure firewall access isn't preventing any issues from the host side? EC uses common HTTPS ports but if you're encountering further issues, you can dig into the Enterprise Connector logs in the installation directory or log a support incident to help investigate.
